Composition** Ingredients Gms / Litre Casein enzymic hydrolysate 5.000 Yeast extract 2.500 Dextrose 1.000 Agar 15.000 Final pH ( at 25°C) 7.0±0.2 **Formula adjusted, standardized to suit performance parameters Directions Suspend 23.5 grams in 1000 ml distilled water. Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. Mix well and pour into sterile Petri plates. Principle And Interpretation Plate Count Agar is formulated as described by Buchbinder et al (1) which is recommended by APHA (2,3,4) and FDA (5). Casein enzymic hydrolysate provides amino acids and other complex nitrogenous substances. Yeast extract supplies Vitamin B complex. APHA recommends the use of pour plate technique. The samples are diluted and appropriate dilutions are added in Petri plates. Sterile molten agar is added to these plates and plates are rotated gently to ensure uniform mixing of the sample with agar. The poured plate count method is preferred to the surface inoculation method, since it gives higher results. Plate Count Agar is also suitable for enumerating bacterial count of sterile rooms. Quality Control Appearance Cream to yellow homogeneous free flowing powder Gelling Firm, comparable with 1.5% Agar gel Colour and Clarity of prepared medium Light yellow coloured clear to slightly opalescent gel forms in Petri plates Reaction Reaction of 2.35% w/v aqueous solution at 25°C. pH : 7.0±0.2 pH 6.80-7.20 Cultural Response Cultural characteristics observed after an incubation at 35 - 37°C for 18 - 48 hours. Ethambutol (EMB, E) is a medication primarily used to treat tuberculosis. It is usually given in combination with other tuberculosis medications, such as isoniazid, rifampicin and pyrazinamide. It may also be used to treat Mycobacterium avium complex, and Mycobacterium kansasii. It is taken by mouth.
Nutrient Agar is a basic culture medium used for maintaining microorganisms (1), for purity checking prior to biochemical or serological testing. It is used for the cultivation and enumeration of bacteria, which are not particularly fastidious.
Sheep Blood Agar Plate MP1301 Used for cultivation of fastidious organisms and studying haemolytic reactions. It provides improved and enhanced haemolysis. Composition** Ingredients Gms / Litre Casein enzymic hydrolysate 14.000 Peptic digest of animal tissue 4.500 Yeast extract 4.500 Sodium chloride 5.000 Agar 12.500 Sheep Blood 5.000 Final pH ( at 25°C) 7.3±0.2 **Formula adjusted, standardized to suit performance parameters Directions Either streak, inoculate or surface spread the test inoculum (50-100 CFU) aseptically on the plate.
Sabouraud Dextrose Agar SMH063 For cultivation of C.albicans in accordance with the harmonized method of USP/EP/BP/JP. Composition** Ingredients Gms / Litre Mycological, peptone 10.000 Dextrose 40.000 Agar 15.000 **Formula adjusted, standardized to suit performance parameters Directions Sabouraud Dextrose Agar is a ready to use solid media in glass bottle. The medium is pre-sterilized; hence it does not need sterilization. Medium in the bottle can be melted either by using a pre-heated water bath or any other method. Slightly loosen the cap before melting. When complete melting of medium is observed dispense the medium as desired and allowed to solidify
Agar is commonly used in the laboratory to help feed and grow bacteria and other microorganisms. It acts as a culture that provides nutrients and a place for these items to grow, but since it is indigestible to the microorganisms, they cannot eat and destroy it.
Agar Powder is specifically produced for use in bacteriological culture media and plant tissue culture media, where clarity and compatibility are not of prime importance. It is used in culture media in following concentrations : For Routine Media: 1.4 to 1.6% ,For Soft Media: 0.5% ,For Semi-solid Media: 0.15%, For Media with Reduced Oxygen Tension: 0.05 - 0.1% ,For Extra Hard Gels, for inhibiting swarming of Proteus species: 2.5% - 3.0% Principle And Interpretation Agar is prepared from species of red seaweeds specially selected for their Agar gel production, using stainless steel equipment, observing good manufacturing practice. It is a Bacteriological grade powder with high mineral / metal content and is advantageous to use in certain media. It is a cream coloured powder having particle size that can pass through 40 ASTM Screen. When suspended in cold water, it swells but does not dissolve. However, it readily dissolves in boiling water and solubility is facilitated by soaking the powder in cold water. Quality Control Appearance Cream coloured powder. homogenous free flowing powder Solubility Freely soluble in hot water at temperatures above 85°C. Insoluble cold water. Clarity A firm solid, clear to slightly opalescent gel is formed at a concentration of 1.5% at 38-40°C. Dye Diffusion Agar dye diffusion :- 18-20mm Reaction Reaction of 1.5% w/v aqueous solution at 25 °C pH : 6.50 - 7.50
